7.Depending on the strain of choice, lysis of bacteria in water might occur. However, this is not an issue for E. coli and Salmonella spp. For instance, preparation of electrocompetent cells through washing with water is a frequently used method [47]. The preparation of bacterial samples can alternatively be performed with 1× PBS (phosphate-buffered saline) or liquid growth medium. Another option is applying a drop of liquid culture.

8.As colony morphology development can differ between plates due to various reasons like moisture or oxygen supply, always apply strains to be compared and the controls onto the same plate.
